IHC-P mouse brain (2 different tumor types) without AR staining: pH 6 and 10 Ab: 1/100, 1/200 (5 ug/ml) - prediluted in the morning, used in the afternoon incubated for 1 h and the /n 2nd: AP , ABC - works fine, otherwise other primaries work fine

I tested this anti-desmin antibody against IHC-Fr in mouse brain tissue, perfused with PFA, post-fixed in PFA for 1 hour, and cryoprotected in PB+sucrose. Stained with different protocols and concentrations (permeabilization with Trinton x 0.1-0.3% in 10% goat serum for 1h and concentrations ranging from 1:50 to 1:1500). I used different non-abcam gt anti-mouse secondary antibodies. I got staining that is not seen in controls without primary antibody, but is not as expected for desmin. It is the staining of the nucleus of some cells and it is supposed to be a cytoplasmic (intermediate filament) staining. He intended to stain blood vessels and pericytes with this antibody. I have an NG2 antibody that stains pericytes and I see them regularly on my stain. This was the first time I had used a desmin specific antibody. I have attached a photograph of the hippocampus (bottom), the corpus callosum (middle), and the deep layers of the cortex (top). It's labeling the pyramidal cell nuclei in the hippocampus, which was actually not expected. I'm sure because there are pericytes in the brain.

Thank you for sending your protocol, which appears to be correct, and the image file. I agree that the antibody is staining something that is probably not desmin. Are you interested in testing a different antibody against desmin? I see that ab15200 was posted in an article that used it to identify pericytes:

Schadler KL et al. The delta-4 ligand plays a pivotal role in the formation of pericytes/vascular smooth muscle cells during vasculogenesis and expansion of tumor vessels in Ewing's sarcoma. Clin Cancer Res 16:848-56 (2010). PubMed: 20103680

Dear Madam or Sir, I would like to request a quote for the following antibodies please: anti-desmin ab6322 antibody anti-desmin ab15200 antibody And I would like to ask your advice on how to choose between these two antibodies. Specifically, I'm going to test human hematopoietic progenitor cells. Do you have any information on which one will give a more specific color in this case? (The immunogen for polyclonal ab15200 is a human synthetic peptide, whereas for monoclonal ab6322 it is a full-length native pig protein). Any information will be greatly appreciated.
